Introduction

The aim of the present study was to investigate the anti-estrogenic and anti-estrogenic effects of an oral estrogen-antagonist in a rat model of breast cancer. Two groups of female Sprague-Dawley rats were assigned to one group (Group A) and the other group (Group B) of female Sprague-Dawley rats which received an oral dose of a 10 mg/kg dose of Tamoxifen citrate (Tamoxifen), which was given as a vehicle. After tamoxifen treatment, the rats were sacrificed. Estrogen-dependent in the liver and prostate were evaluated by the in vitro testosterone-dependent testosterone assay. Materials and methods

Animals

The present study was a single center animal study conducted at our institution from July 2014 to September 2015. The rats were purchased from the local animal house at Nanyang University (Changzhou, China) and were randomly divided into two groups (Group A and B) which received Tamoxifen (10 mg/kg, i.p.) or vehicle (nolvadex, a vehicle) at a dose of 10 mg/kg. The two groups were maintained under the same conditions, respectively, for a period of 7 days. The rats were acclimatized for 5 days and received a single intraperitoneal injection of tamoxifen (10 mg/kg) orally in a drinking water (pH 5). After 1 week, the rats in the Group A received 10 mg/kg tamoxifen (nolvadex) or vehicle. After this, the rats in the Group B received tamoxifen (10 mg/kg) orally. The rats in both groups were randomly assigned to receive an oral dose of Tamoxifen 10 mg/kg orally (nolvadex) or vehicle (nolvadex) every other day. The rats in the treatment groups were kept under the same conditions as in the control groups for a period of 7 days. All the experimental procedures were approved by the Ethical Committee of Nanyang University.

Treatment groups

All the experimental procedures were performed in accordance with the approved protocols in accordance with the National Institutes of Health Guide for the Care and Use of Laboratory Animals.
